Objectives |

The course aims at studying the basic techniques of the DNA cloning using the plasmid of Escherichia coli.
|

Outline |
|
The purpose of this course is to learn the following techniques
(1) The ligation of the lac DNA and the transformation of E.coli.
(2) The purification and identification of the recombinant plasmid DNA
(3) Restriction mapping
(4) In vitro amplification of the lac DNA with PCR
(5) The induction of the lac gene and the measurement of the β-galactosidase
activity
